Derivation of Reaction Rate Expression with Enzyme Inhibition

Biochemical Engineering

Enzyme inhibition can be classified into the following three categories, depending on the mechanism.

  1. Competitive
... The inhibitor binds to the active site and competes with the substrate.
  2. Non-competitive
... The inhibitor binds to a differen site and reduces enzyme activity.
  3. Un-competitive
... The inhibitor binds to and inactivate the enzyme-substrate complex.
Derive the reaction rate expression with equilibrium assumption. Find the effect of different levels of inhibitors on the regular velocity-versus-substrate plot and the Lineweaver-Burk double reciprocal plot.
