METHODS

A container of plain Yoplait yogurt was obtained. The label stated that the ingredents included Lactobacillus Acidophilus cultures. The yogurt was swabbed onto plates containgLactobacillus Selection Agar. The plates were then placed into an anaerobic chamber containing a gas pack and incubated at 37 degrees Celsius. After 48 hours the plates wereexaminedfor growth. Isolated colonies from the plates were restreaked usingthe streakplate method. These plates were incubated under same conditions. New plates were then observed macroscopically. Those plates with coloniesthat showed the same characteristics as outlined in our research were further analyzed using the following tests:
 
 

    1. Gram stain
    2. Endospore stain
    3. Plated on MacConkey’s
    4. Motility stab
    5. Catalase test
    6. Lactose fermentation test
    7. Glucose fermentation test
    8. Incubation outside of anaerobic chamber (in presence of oxygen)
