Results
Streak plates of colonies isolated from soil inoculated
plates are shown in figures one and two. A positive control for Streptomyces
is presented in figure three. Gram stains of 5 selected colonies from the
streak plates that exhibited characteristic Streptomyces morphology are
shown in figures 5-10. Both plates exhibited the characteristic earthy
smell of Streptomyces. Figure four is a positive control for Streptomyces
Gram stain. All colonies selected for Gram stain tested catalase positive.
All five samples proved to be mixed cultures and only one appeared to possibly
contain Streptomyces (figure 8).
Colonies:

Gram Stains:
*All pictures were taken under a magnification of 1000X, using oil
immersion.

Fig 5: Sample 1 is a mixed culture of gram positive and gram negative organisms.
As compared to the control, these organisms are much larger, and differ
in shape.
Fig 6: Sample 2 is also much larger and differ in shape.
Fig 7: This slide of sample 3 shows the presence of a gram positive,
spore forming organism.
Fig 8: This is also of sample 3, so this is most likely a mixed culture
of gram positive and gram negative spore forming organisms.
Fig 8: Sample 4. This is clearly a mixed culture, but the highlighted bacteria
show similar shape to the control, and could possibly be Streptomyces.
Further isolation would be necessary. There are other gram negative and
gram positive bacteria present in this culture.
Fig 9: This is also sample 4. This is a more condensed image of the slide,
showing a broader range of organisms present in the culture. One can also
see spores, and endospores on this slide.
Fig 10: Sample 5. Again, this is a mixed culture. There are endospores
and spores present, indicating spore forming organisms. There are also
both gram negative and gram positive bacteria on the slide.
